"Proteasome Function and Distribution: From Ubiquitin-Independent Degradation and Phase Separation to Neuronal Visualization"

Proteasomal degradation is central to maintaining protein homeostasis, traditionally via the ubiquitin–proteasome system (UPS). We uncovered that the 20S proteasome can also degrade intrinsically disordered proteins independently of ubiquitin and ATP. Using CRISPR-based fluorescent tagging, we tracked the live dynamics of 20S and 19S complexes in cell lines, embryonic stem cells, and differentiated neurons. This talk will highlight the distinct behaviors of proteasome complexes under osmotic stress and in neuronal contexts.
